Electronic nose knows when your cantaloupe is ripe

ScienceDaily (Mar. 30, 2012) ? Have you ever been disappointed by a cantaloupe from the grocery store? Too ripe? Not ripe enough? Luckily for you, researchers from the University of California, Davis might have found a way to make imperfectly ripe fruit a thing of the past.

The method will be published on March 30 in the Journal of Visualized Experiments (JoVE).

"We are involved in a project geared towards developing rapid methods to evaluate ripeness and flavour of fruits," said paper-author Dr. Florence Negre-Zkharov. "We evaluated an electronic nose to see if it can differentiate maturity of fruit, specifically melons. The goal is to develop a tool that can be used post-harvest to better evaluate produce, and develop better breeds."

When fruit ripens, it develops a characteristic volatile blend, indicating its maturity. Traditionally, the gold-standard of evaluating these volatiles has been gas chromatography, but it takes up to an hour to analyze a single sample, which makes it impractical to use outside the lab. Dr. Negre-Zakharov and her team wanted to determine if the much cruder -- but much faster -- electronic nose was able to determine if the melon they used in the experiment were ripe. It was.

"It's quite encouraging technology for the purposes of determining maturity," she said.

The project is part of the Specialty Crops Research Initiative, funded by the United States Department of Agriculture, which was "established to solve critical industry issues through research and extension activities." Dr. Negre-Zkharov and her team are working on quantitative methods of evaluating fruit-ripeness in the hopes that it will help the industry produce better quality produce.

"It's very impressive that the electronic nose system can do a type of gas chromatography in about a minute. Ultra-fast, indeed. Also, the sample preparation is as easy as making a smoothie at home. Such a user-friendly system could greatly help analysis efficiency in this field," said JoVE Science Editor, Dr. Zhao Chen. "Given the popularity of JoVE video-articles, I expect many researchers will know and adopt this method in their own research."

Since the very nature of the project is to give people useful tools, the researchers decided to publish in JoVE, the only peer reviewed, PubMed-indexed science journal to publish all of its content in both text and video format.

"We thought that the best way to get people to adopt the method was showing a video, instead of publishing a text," said Dr. Negre-Zkharov.

The next step is to take the electronic nose out into the field to determine if it can still determine fruit maturity with all of the background smells interfering -- like soil and air-quality. Though the team has already tested the device in the field, they have not yet analyzed their results.

'Earth Hour' Pauses at U.S. Border

Consider an hour without power, from 8:30 to 9:30 p.m. Saturday, local time. Organizers say as many as 1.8 billion will join in the symbolic environmental event worldwide. But if you live in the US, your neighbors may think you just blew a fuse. ?

Earth Hour starts in the mid-Pacific Islands on Saturday night, March 31 with an hour of voluntary electrical blackouts in Tonga. ?

The event moves on to the western Pacific and Indian Oceans, where stars helping promote the event include an Australian model, an Indian cricket champion and a Russian heavyweight boxer.?

Following the sunset, Earth Hour heads west, where hotels and office buildings across the Persian Gulf will go dark. In Dubai, the world?s tallest building ? the Burj Khalifa ? will stand lightless. Five-star hotels in Saudi Arabia will burn a few hundred gallons less oil in that quiet hour. ?Even Libya, fresh from many involuntary blackouts, will host Earth Hour events.?

A long list of events are also scheduled for Europe: Big Ben, the Eiffel Tower and the Leaning Tower of Pisa will all go dark ?for one hour when the clocks strike 8:30 p.m.?

Barely notice
Yet few Americans have ever heard of Earth Hour, and only a few dozen high profile events are scheduled. While the Empire State Building and the United Nations in New York City will go dark, most of New York will barely notice.

Earth Hour has only one US press coordinator to publicize the event. Slovenia has two. India has three. Brunei has five.?

Conceived in Australia in 2007 at World Wildlife Fund's Sydney office, and promoted by the Sydney Morning Herald newspaper, Earth Hour?s global popularity may reflect a disparity between U.S. and international receptivity to environmental symbolism. ??

?In Asia and the Middle east, you'll find that people are less aware about the risks of climate change and global warming,? a spokesman for the Kuwaiti team said. ?The Earth Hour campaign carries a simple message ? so that people can be easily engaged environmentally.??

Of course, the U.S. has had its own original Earth Day celebration since 1970. A Google news search delivers 33,000 links focused on Earth Day, more than three weeks away. Earth Hour, meanwhile, had only 6,000 hits hours before it began in the far Pacific.?

Strikes a chord
The original Earth Day has been the centerpiece of the US and European environmental movement for four decades, and it continues to engage grassroots groups around world, said Dennis Hayes, the principal ?Earth Day founder who continues to chair the global advisory committee.

The lack of enthusiasm stateside for Saturday's event could be cultural: From Earth Day to Earth Hour seems like downsizing, a concession to the less committed, Grist Magazine joked.?

But the serious news about Earth Hour is nearly all international.

Hundreds of items in small publications in Asia, Africa and the Middle East show that the event strikes a chord in smaller cites as much as large ones, and in traditional societies as much as westernized ones.?

Typical was a statement by the mayor of Taguig ? a city of 600,000 in the Philippines. "Participating in the activity signifies one's support for environmental protection, as well as the campaign to solve or at least mitigate the ill effects of climate change to the environment," said Lani Cayetano.

So, who are these food writers?

One of them is Joe Yonan. He's the food and travel editor of The Washington Post (formerly at the Boston Globe). The team at the Post has twice been awarded the James Beard Foundation award for the nation's best newspaper food section. He also writes features for both Food and Travel, including the monthly "Cooking for One" column, which won honors from the Association of Food Journalists. He's the author of "Serve Yourself: Nightly Adventures in Cooking for One," from Ten Speed Press, and the coauthor of "The Fearless Chef" with Boston chef Andy Husbands. I am the restaurant reviewer/food writer for the Portsmouth Herald and author of "Maine Classics: More than 150 Delicious Recipes from Downeast," which I wrote with Mark Gaier and Clark Frasier from Arrows Restaurant. I'm not sure what I'll be reading yet, but you'll certainly get to sample dishes from recipes in the book. I'll likely mix in some columns and samples from my novel in progress, which has a heavy food element.

As each author reads, guests will savor and sample prepared dishes from recipes in the author's books. One of them is Joe Yonan. He's the food and travel editor of The Washington Post (formerly at the Boston Globe). The team at the Post has twice been awarded the James Beard Foundation award for the nation's best newspaper food section. He also writes features for both Food and Travel, including the monthly "Cooking for One" column, which won honors from the Association of Food Journalists. He's the author of "Serve Yourself: Nightly Adventures in Cooking for One," from Ten Speed Press, and the coauthor of "The Fearless Chef" with Boston chef Andy Husbands. ScienceDaily (Mar. 27, 2012) ? The novel H1N1 flu pandemic in 2009 underscored weaknesses in methods widely used to diagnose the flu, from frequent false negatives to long wait times for results. Now a four-year, National Institutes of Health-funded study of 146 patients with flu-like symptoms spearheaded by Associate Professor Catherine Klapperich (BME, MSE) has validated a prototype rapid, low-cost, accurate, point-of-care device that promises a better standard of care. Once optimized and deployed in the clinic, the new device could provide clinicians with an effective tool to quickly diagnose both seasonal and pandemic strains of influenza, and thus limit the spread of infection.

The study's research team -- Klapperich, Qingqing Cao(ME PhD'11), Madhumita Mahalanabis(BME postdoctoral fellow), Jessie Chang (BME MS'10), Brendan Carey (BME'11), Christopher Hsieh (BME'11) and Ahjegannie Stanley (summer intern) from the College of Engineering; medical personnel from the Boston University Medical Center (BUMC) Emergency Department; and an infectious disease physician from Beth Israel Deaconess Medical Center (BIDMC)/ Harvard Medical School -- published its findings in the March 22 online edition of PLoS ONE.

To produce a faster, cheaper, highly accurate flu diagnostic test that could be run at the point of care, the researchers miniaturized an expensive, three-hour, lab-scale diagnostic test -- known as RT-PCR and now considered the gold standard in flu detection -- into a single-use microfluidic chip. About the size of a standard microscope slide, the integrated chip consists of a column at the top that extracts RNA from signature proteins in the sample associated with the influenza A virus; a middle chamber that converts the RNA into DNA; and a climate-controlled lower channel used to replicate the DNA in sufficient quantities so it can be detected by an external reader.

Working with two types of nasal specimens, the researchers used the chip to produce results that matched the high accuracy and relatively fast turn-around time of the lab-scale method.

"We wanted to show that our technique was feasible on real-world samples prepared on the chip," said Klapperich. "Making each chip single-use decreases the possibility of cross-contamination between specimens, and the chip's small size makes it a good candidate for true point-of-care testing."

The microfluidic chip also proved far more effective than other commonly used flu diagnostic tests including viral culture, a lab procedure requiring up to a week to produce results; rapid immunoassays, which work like pregnancy tests but were only 40 percent reliable in detecting the presence of a flu virus in this study; and direct fluorescent antigen testing (DFA), a more accurate but labor-intensive process in which medical personnel prepare and interpret samples stained with fluorescent antibodies.

"The new test represents a major improvement over viral culture in terms of turn-around time, over rapid immunoassay tests in terms of sensitivity (ability to detect the virus from minimal sample material) and over DFA and RT-PCR in terms of ease of use and portability," Klapperich observed.

Ultimately seeking to enable clinicians to use their microfluidic chips for frontline flu virus detection, the researchers next plan to optimize their method so that it can produce results in a third less time (an hour) with chips that cost half as much to make (five dollars). In addition, they are exploring ways to develop a lower cost external reader that's no bigger than a clinical digital thermometer.

NASA Hopes to Launch 5-Rocket Mission to Light US East Coast Sky Tuesday

NASA will try again early Tuesday (March 27) in an effort to launch five suborbital sounding rockets on a mission to study high-level jet stream winds by creating artificial glowing clouds near the edge of space.

After several delays, the rockets are scheduled to blast off from launch pads in Virginia between 2 a.m. and 5 a.m. EDT (0600 and 0900 GMT).

If all goes well, each of the five unmanned rockets will release a chemical tracer into the jet stream winds more than 60 miles up that should create brilliant milky white?trails in the nighttime sky?to enable scientists and people on the United States East Coast to actually "see" these?high-altitude winds at the edge of space, NASA officials have said.

The glowing clouds created in the experiment may also be visible to skywatchers along the U.S. East Coast, local weather permitting.

But while the latest weather forecasts appear to be favorable with clear weather forecast along the Atlantic Seaboard, another meteorological condition ? gusty winds ? could ground the rockets for yet another night. The mission, which is called the?Anomalous Transport Rocket Experiment, or ATREX for short, has been on hold since the night of the first launch attempt on March 15.

That night, radio frequency interference on one of the five rockets precluded a launch. Since then, unfavorable weather has been the main problem. In addition to the launch site, NASA has also set up cameras to photograph the?rockets' luminous vapor trails?from two locations: the Rutgers Marine Field Station in Tuckerton, N.J., and the U.S. Army Corps of Engineers at Duck, N.C. Clear skies at all three locations are essential in order for the rockets to be launched.?

There are several ways to watch NASA's ATREX mission launch overnight:

Promising early attempts thwarted

On Thursday (March 22), mission scientists looked like they were finally going to get their chance to launch, with mainly clear skies and only a few scattered high clouds at the North Carolina viewing site.

The three-hour launch window was set to open at midnight EDT (0400 GMT), but as countdown clocks ticked closer to the scheduled opening of the launch window, patchy areas of low clouds as well as fog began to creep into Wallops Island and Tuckerton, N.J. Occasional gusty winds at the launch site also posed a problem.

When the launch window opened at midnight, early Friday morning, the five?sounding rockets?were still on their respective launch pads. NASA continued to watch the weather, hoping that clearer skies might evolve as the night progressed, but in his periodic briefings, NASA's weather officer could only report that local conditions were in fact, slowly worsening.

To add to NASA?s problems, it was discovered that three boats had apparently wandered into the restricted zone, located immediately offshore from the Virginia launch site.

Two of the boats immediately moved out, but the last boater refused to leave the restricted waters which are potentially dangerous because the rockets could land there.

At 1:30 a.m. EDT (0530 GMT) this morning, it was determined that it would take a minimum of one hour to move the final boat out of the restricted area. With low clouds and fog continuing to close in at both Wallops Island and Tuckerton, the decision was made a minute later to scrub the mission for the night.?

Launch night decisions

A sharp cold front moving off the Mid-Atlantic coast early today is expected to bring in a sweep of dry and chilly Canadian air promoting clear and starry skies for overnight tonight. However, northerly winds gusting as high as 30 mph at the launch site could pose another problem, either delaying or postponing the launch yet again. [In Images: Mysterious Night-Shining Clouds]

So NASA managers have now decided to wait until no earlier than 2 a.m. EDT (0600 GMT) on Tuesday in hopes that the expected strong winds will either have abated or at least diminished enough to allow for a launch.? The launch window will remain open for three hours.?

If the launch is held over until Tuesday night/Wednesday morning the forecast show? that the wind will have switched direction and blow much lighter ? south-southwest at only around 5 mph. But there will then be a concern about a swath of mid-to-high level clouds that could drift in from the west by launch time.? On Wednesday morning the window is scheduled to open at 12:17 a.m. EDT and will close at 4:55 a.m. EDT (0417 to 0855 GMT).

The ATREX mission ? when it finally launches ? will?provide an interesting sky show?that might puzzle and amaze unsuspecting viewers along the United States East Coast, from central New England to South Carolina, and as far west as West Virginia.

Each rocket will eject a chemical tracer, called trimethyl aluminum (TMA), which will be illuminated at high altitudes by the sun (which will be below the local horizon at ground level).?Initially, the cloud trails ? seven in all ? are expected to glow in?reddish hues, and then quickly turn to white. They could persist in the sky for possibly as long as 20 minutes before fading completely away. The launch window runs from midnight to 3 a.m. EDT (0400 to 0700 GMT).

How it happens

Trimethyl aluminum is an organometallic compound that is liquid at room temperature. Though nontoxic, TMA is dangerous because it can ignite spontaneously.

In rockets, the chemical is kept under pressure. At the very low pressures of the?Earth?s upper atmosphere, some of the released TMA evaporates, but the rest freezes into solid particles. At an altitude of roughly 62 miles (100 kilometers), heating by friction causes further evaporation, and the particles shine in part by a chemiluminous process.?

In several cases, bluish-white cloud "puffs" will be created, resulting from the chemical reaction of TMA with atomic oxygen in the atmosphere, producing the molecule AlO. While NASA's ATREX launch will occur in the middle of the night, at the high altitude where the TMA will be released, it will be directly illuminated by sunlight, which will excite the molecules causing them to glow.

What do we learn?

Using time-lapse photography from the New Jersey and North Carolina sites, the drift of the cloud trails can be accurately determined, which indicate the direction and?speed of the winds?at these high altitudes.?

Such winds continually transfer heat and matter from the Earth?s sunlit hemisphere around to its dark side. In addition, the rate of diffusion of the trails will give scientists an idea of how fast the TMA that was deposited will disappear.

The rate of diffusion is inversely proportional to the density of the air which decreases in height. With this in mind, the cloud trails and puffs that are released at higher altitudes will likely appear wider and larger, and should expand much faster than those trails that are released at lower altitudes.

Stocks rally at open, rebound from slide

U.S. stocks were higher Monday, rebounding after the S&P's worst weekly performance so far this year, as investors looked ahead to data on February pending home sales.

The Dow Jones industrial average was up 100 points in the early going, while the Nasdaq Composite index touched a multi-year high.

The benchmark S&P 500 index fell 0.5 percent last week, a relatively minor decline that was still the biggest weekly slide since the final week of December.

Rising optimism about economic growth has kept investors piling into equities, a trend that looked set to continue with the S&P 500 near its highest point since May 2008. The Nasdaq has risen for six straight weeks.

"There's a lot of optimism in the market, and the longer-term trend is up," said Cort Gwon, chief strategist at HudsonView Capital Management in New York. "After a quiet weekend we'll probably just feed off the past few weeks, which were largely very positive."

Crude oil prices fell 0.1 percent, though they remained up almost 8 percent for the year on worries about supply disruptions from the Middle East. Rising prices are viewed as a headwind for the economy.

Pending home sales are seen rising 1 percent in February after a 2-percent increase in January. Data on the housing market continues to be weak, with the government reporting last week that new home sales fell 1.6 percent. The pending home sales data will be released at 10 a.m. ET.

Lions Gate Entertainment Corp rallied 8.9 percent to $15.83 after the strong opening to its film "The Hunger Games," which made $214 million over the weekend globally.

Cal-Maine Foods Inc reported third-quarter earnings that fell from the prior year on rising feed costs, which the company sees persisting through the summer. Apollo Group Inc is scheduled to report later Monday.

BATS Global Markets Inc on Sunday apologized for a system failure that caused its own shares to erroneously trade for less than a penny on Friday and resulted in Apple Inc's shares being temporarily halted.

Chief Executive Joe Ratterman called the incident a "devastating moment," and said the company's withdrawn initial public offering was on hold "for the foreseeable future."

U.S. stocks rose in light volume on Friday, buoyed by rising energy and basic materials shares.
