STANFORD, Calif. Stanford University School of Medicine scientists have demonstrated, in a study conducted jointly with researchers at Yale University, that induced-pluripotent stem cells the embryonic-stem-cell lookalikes whose discovery a few years ago won this year's Nobel Prize in medicine are not as genetically unstable as was thought.

The new study, which will be published online Nov. 18 in Nature, showed that what seemed to be changes in iPS cells' genetic makeup presumed to be inflicted either in the course of their generation from adult cells or during their propagation and maintenance in laboratory culture dishes instead are often accurate reflections of existing but previously undetected genetic variations among the cells comprising our bodies.

That's good news for researchers hoping to use the cells to study disease or, someday, for regenerative medicine. But it raises the question of whether and to what extent we humans are really walking mosaics whose constituent cells differ genetically from one to the next in possibly significant respects, said Alexander Urban, PhD, assistant professor of psychiatry and behavioral sciences. Urban shared senior authorship of the study with bioinformatics professor Mark Gerstein, PhD, and neurobiology professor Flora Vaccarino, MD, both of Yale.

It's only a few years ago that human iPS cells started becoming available to researchers. These cells hold great promise because they act almost exactly like embryonic stem cells, which can be nudged to differentiate into virtually any of the body's roughly 200 different cell types. But iPS cells can be derived easily from a person's skin, alleviating numerous ethical concerns arising from the necessity of obtaining embryonic stem cells from fertilized eggs.

At least in principle, iPS cells' genetic makeup closely reflects that of the individual from whom they were derived. Today, "heart cells" derived from a heart patient's skin can be produced in a laboratory dish so scientists can learn more about that particular patient's condition and to screen drugs that might treat it. Tomorrow, perhaps, such cells could be administered to that patient to restore heart health without being perceived as foreign tissue by the patient's immune system, which would otherwise reject the implanted cells.

However, Urban said, several previous studies have raised worries regarding iPS cells' genomic stability. Whether it was the reprogramming procedure researchers use to convert ordinary adult cells into iPS cells or the culturing techniques employed to keep them alive and thriving afterward, something appeared to be inducing an upswing in these cells' manifestation of copy number variations, or CNVs the disappearance or duplication of chunks of genetic material at specific locations along the vast stretches of DNA that coil to form the chromosomes residing in all human cells.

CNVs dot everybody's genomes. They occur naturally because of DNA-copying errors made during cell replication, and accumulate in our genomes over evolutionary time. The human genome, taken as a whole, is a DNA sequence consisting of four varieties of chemical units, strung together like beads on a roughly 3-billion-bead-long necklace. CNVs range in length from under 1,000 DNA units to several million. They account for up to several percent of the entire human genome, making them a major source of genetic differences between people.

But if either iPS cells' mode of generation or their subsequent maintenance in culture were promoting an increase in CNVs, it would seriously compromise these cells' utility in research and pose a fatal flaw to their use in regenerative medicine, said Urban. "You would never want to introduce iPS cells into a patient thinking that these cells had the same genome as the rest of the patient's cells, when in fact they had undergone substantial genetic modifications you knew nothing about, much less their effects," said Urban. (Similar concerns apply to embryonic stem cells.)

To see how serious a problem CNVs might pose for iPS cells' use, the collaborators performed tiny skin biopsies on seven volunteers and extracted cells called fibroblasts, which abound in skin and are amenable to cell culture in general and iPS cell generation in particular. From these, the team produced 20 separate iPS cell lines in culture. Using now-standard lab methods, the investigators determined, chemical unit by chemical unit, the full genomic sequence of the cells composing each new iPS cell line.

Urban and his colleagues, who had likewise assessed the fibroblasts from which the lines were derived, compared their genomic sequences with those of the newly generated iPS cells. The scientists were able to pinpoint numerous CNVs in the new cells that hadn't shown up in the fibroblasts. This raised the possibility that the rigors of reprogramming or life in a dish, or both, had led to new CNVs in the cells.

But the technique used to determine the full-length genome sequences of iPS cells and the "parental" fibroblasts from which they'd been spawned analyzes not single cells but millions at a time. So a CNV residing in only a minority of cells within this mix could easily be missed, its signal swamped by the noise of the majority report.

Armed with knowledge of the precise locations along the genome where each of the "new" CNVs had popped up in their iPS cell lines, the scientists went back to the fibroblasts. This time, they used an analytical tool that, like a photocopying machine, can generate millions or billions of copies of a single section of DNA provided that the specific DNA section is present to begin with. For each "new" CNV that had been unearthed in the iPS cells, a different version of this molecular copying machine was employed.

"Lo and behold, in many cases, this technique unearthed CNVs in the fibroblasts that were there all along but had been missed in the earlier, mass analysis," Urban said. These CNVs had gone undetected because the fibroblasts in which they resided represented as little as a fraction of a percent of the fibroblasts in a biopsy sample. But any CNV in a fibroblast lucky enough to become the "parent" of the billions of iPS cells bearing its identical genomic "face" would now stick out like the proverbial sore thumb. Six out of the 20 different iPS cell lines sported at least one CNV unshared with the other lines, the investigators found. To the extent this reflects differences in parental fibroblasts, it means about 30 percent of the cells in human skin may harbor unique CNVs.

"This is a huge amount of cell-to-cell genetic variation," said Urban, who also participated in a recent study of human tissues conducted by the laboratory of Michael Snyder, PhD, professor and chair of genetics at Stanford. That study showed there can be distinct CNV patterns in kidney, pancreas and liver. "Rather than monoliths, our bodies may be mosaics composed of cells whose genomes differ. What we do not know is whether or when these differences are dangerous, irrelevant or beneficial."

That may depend on the organ, said Urban. "The more complicated something is, the more ways there are that something could go wrong with it. The brain is a particularly complicated organ. CNVs affecting cells in particular brain structures or areas could be playing a role in complex neurodevelopmental disorders such as schizophrenia and autism." Urban intends to find out, he said.

Lane Kiffin says he will be back as USC's football coach

USC's Lane Kiffin has told reporters at the Rose Bowl that he has been assured he would return as the Trojans' football coach.

Kiffin's comments came in the wake of a 38-28 loss to UCLA on Saturday afternoon that made the Bruins champions of the Pac-12 South Division and gave them a spot in the conference title game.

USC opened the season ranked No. 1 in the Associated Press media poll, but the Trojans have lost four Pac-12 games -- to Stanford, Arizona, Oregon and UCLA.

The Trojans came into Saturday's game ranked No. 21 and could fall out of the top 25.

While his team has not performed up to expectations, Kiffin has found controversy off the field. He lied about voting USC No. 1 in a poll early in the season, became embroiled in a tiff about media access and the false accusation of a reporter who wrote about an injury, ruffled feathers by changing the numbers of players during a blowout win over Colorado and most recently denied he had anything to do with a student manager deflating footballs below regulation level during a loss to Oregon.

His father, Monte, has also been criticized for the peformance of USC's defense in losses to Arizona and Oregon.

After Saturday's game, UCLA Coach Jim Mora credited Lane Kiffin as a "great play-caller" and Monte Kiffin for having "one of the great defensive minds" in football.

Is Bradley Cooper still the sexiest? Mais oui!

Bradley Cooper may have passed People magazine's Sexiest Man Alive torch to successor Channing Tatum earlier this week, but Cooper doesn't need an official sexy title to prove his appeal. All he has to do is speak ? in French.

OK, sure, the actor's English is pretty swoon-worthy too, but when the Cooper switched to his second language during his Friday morning visit to TODAY, there was no denying that it works for the star.

And as if speaking the language of love wasn't enough, Cooper also pulls out all the stops in his new film, "Silver Linings Playbook," by showing off his ballroom dancing moves alongside co-star Jennifer Lawrence.

Waltzes and tangos aside, his character in "Playbook," Pat, isn't quite as charming as Cooper is in real life. After all, Pat struggles to cope with anger and day-to-day life after leaving a mental institution. Still, despite the obvious differences, Cooper's own mother saw some parallels between her son's off-screen persona and the one on-screen.

"He's got anger management issues and a song can trigger a violent episode -- yeah, it's very autobiographic," he joked. "I took that (comparison by my mom) harshly, I've got say, because she meant it. I walked downstairs after we did a very intense scene and that's when she said, 'Bradley, it's like you're not acting.' And I thought, 'What does that say?!'"

Combat City USA Lets Customers Shoot Each Other

A Florida gun range called Combat City USA has taken the idea of target practice to a completely different level and is now allowing customers to shoot real guns at each other in simulated combat.

But don?t worry, you can?t be killed by the guns (at least not how they allow them) because they have been modified into airsoft guns, reports Yahoo! News. Owner Dave Kaplan stated:

?A revolver would work the same, semi auto, the action actually cycles. There?s full recoils with the weapon. The assault weapon fully cycles.?

While the airsoft pellets won?t inflict serious harm, they do hurt. A user on the Combat City website explains:

?There is a degree of pain associated with airsoft just like paintball. It is significantly less than paintball and without the swelling.?

For added safety during the simulated combat, participants are outfitted with protective gear including helmets and padding over sensitive areas. Kaplan explained:

?There is supposed to be a degree of pain so that you do learn from it. Someone?s trying to hurt you. You learn how tactical as a civilian you can be.?

NBC Sports notes that Tiffany Chapin, a single mom, believes the real-life scenarios she creates at the gun range are the best way for women to learn how to defend themselves. Chapin teaches classes specifically for women who want to learn self-defense. Kaplan stated of the women, ?Most of the time, they feel a little bit uneasy, nervous. I think it helps to have a female instructor.?

Combat City was created in the space that was once occupied by a grocery store. It has been modified to become an indoor combat setting. Customers are broken up into teams and play games like capture the flag or participate in hostage simulations. As for age limit? The company?s website says, ?all ages are welcome.? They add:

?We can not tell you what you or your child can handle. There are young kids playing at Combat City on a daily basis, only you can decide.?

While each scenario at Combat City lasts between 10 minutes and an hour, customers are allowed to stay the entire day. The cost to get into the firing range is $150, which includes the cost of modifying the gun.

BBC crisis: Wronged politician to agree legal deal

A senior UK politician wrongly implicated in a child sex abuse scandal by the BBC, is likely to reach a legal settlement soon and may sue individuals who named him on Twitter, his lawyer said.

Lawyer Andrew Reid told the BBC that a deal between Alistair McAlpine and the broadcaster was expected later Thursday.

The BBC has already apologized for linking McAlpine, a member of the House of Lords, to child sex abuse that happened decades ago in Wales. The mistaken report, broadcast nearly two weeks ago, has caused turmoil within BBC management ranks and led to the resignation of its chief.

McAlpine, 70, told BBC radio he had been shocked by the report, which did not directly name him but led to Internet chatter about his purported role.

BBC Director General George Entwistle resigned on Saturday as the BBC spiraled further into scandal over its coverage of two separate sex abuse cases ? one, a cover up, and the other, a possible wrongful accusation. NBC's Keir Simmons reports.

He said the BBC had not contacted him to try to verify the report before it was televised on its "Newsnight" program. He said he would have told the broadcaster the reports were false.?

"They should have called me and I would have told them exactly what they learned later on ? that it was complete rubbish," he said.

He expressed sympathy for the sex abuse victim who had mistakenly told BBC that McAlpine was the culprit, pointing out that the victim had suffered greatly because of the abuse.

"But it wasn't me," McAlpine said.

He said some of the damage done by the false report could never be undone.

"It can be repaired to a point," McAlpine said. "But there is a British proverb, which is insidious and awful, where people say 'there's no smoke without a fire' ? you know, 'he appears to be innocent, but...'

McAlpine said "to find yourself a figure of public hatred, unjustifiably, is terrifying."

Among those who could face legal action for identifying McAlpine as the un-named figure in the ?Newsnight? report is Sally Bercow, wife of the Speaker of the House of Commons.

His lawyer advised people who had named McAlpine on Twitter to come forward or face expensive lawsuits.?

"It's easier to come forward and see us and apologize and arrange to settle with us because, in the long run, this is the cheapest and best way to bring this matter to an end," Reid said.

The BBC, which was already facing severe criticism for its handling of child sex abuse claims against its late TV host Jimmy Savile, has broadcast a complete on-air apology for the erroneous investigative report into historic child abuse in Wales.

Meanwhile, police on Thursday arrested Dave Lee Travis, a former BBC radio DJ and television host, as part of the widening investigation into historic allegations of sex offences that began with complaints about Savile.

Police said Travis, who they did not officially name but whose identity was confirmed to ITV News by neighbors, was held on suspicion of sexual offences and taken into custody. The BBC later said it was dropping a scheduled broadcast of an archive episode of music show Top of the Pops, dating from the 1970s, which Travis had hosted.

Police are dividing their investigation into three strands: offences allegedly committed alone by Savile, who died last year aged 84, offences committed by him with others and offences committed by others alone.

The statement said Thursday's arrest fell into the third category, "others." It was the fourth arrest in an operation codenamed "Yewtree".

If you meet multiple people during a job interview, who should receive your?thank you note?

The correct answer is to send a note to every one you meet, said a report published today on TheLadders.?

And make sure the content in each note is different.

"Tailor each communication by focusing on the most significant points raised during each interview,"?Amanda Augustine, job search expert for TheLadders, said in the survey.

"Don?t be afraid to mention little details you learned about the interviewer, such as a shared passion or an upcoming trip ? this demonstrates your attention to detail and makes the message more memorable."

You should aim to send the thank you note via email within 24 hours. However, keep the company's culture in mind because if you're interviewing with a more traditional organization, they may?prefer a hand-written letter?instead.

But whatever you decide to do, just remember to send it.

"Choosing not to send a thank you note jeopardizes your positive perception as being lazy, ungrateful or disinterested," Augustine said.??

BELIZE CITY - Computer security industry pioneer John McAfee says he has gone into hiding in Belize because he believes authorities there are trying to frame him in the slaying of a neighbor, a crime he says he did not commit, according to Wired magazine.

Belize police are searching for McAfee as "a person of interest" in a murder investigation.

"You can say I'm paranoid about it, but they will kill me, there is no question. They've been trying to get me for months. They want to silence me," Wired quoted McAfee as saying on its website. "I am not well liked by the prime minister. I am just a thorn in everybody's side."

The magazine reported that McAfee, 67, contacted one of its reporters by telephone after his neighbor Gregory Faull, was found dead on Sunday in a pool of blood. The 52-year-old American was apparently shot in the head.


Police say McAfee had a history of conflict with Faull.

According to Wired, Faull and other neighbors had been complaining about a half-dozen dogs that McAfee kept at his beachside compound. Wired reported that McAfee sent an email to a reporter Friday night saying his dogs had been poisoned that night and alleging that Belize authorities were responsible.

The reporter said that he spoke to McAfee on Saturday morning and McAfee said he didn?t believe any of his neighbors would poison his dogs. Wired quoted McAfee speaking of Faull: ?This is not something he would ever do. I mean, he?s an angry sort of guy but he would never hurt a dog.?

Faull was found dead on Sunday with a gunshot wound to the back of his head.

McAfee, who amassed a fortune by building the anti-virus company that bears his name, has homes and businesses in the Central American country.? Police say he has lived for at least two years, but Wired said he moved there in 2008.

It was not the first time McAfee, who has tattoos, a goatee and moustache, and a penchant for guns, has drawn police attention.

His premises were raided earlier this year after he was accused of holding firearms, though most were found to be licensed. The final outcome of the case is pending.

He was also suspected of running a lab to make the synthetic drug crystal meth.

American businessman Gregory Faull, seen in August, was found dead at his homee in Belize.

"He was suspected (of making crystal meth) but he was not convicted nor was he charged. He was only suspected," said Belize police spokesman Raphael Martinez.

McAfee also owns a security company in Belize as well as several properties and an ecological enterprise.

Reuters has been unable to reach McAfee, who police want to question.

"It would be quite nice for him to come in and answer some of the questions that could lead to the closure of this case," Martinez said. "He is not wanted for murder, but he is wanted for questioning as a person of interest."

The Belize police department has reached out to counterparts in neighboring Mexico and Guatemala, asking them to detain McAfee if he leaves Belize overland.

McAfee was one of Silicon Valley's first entrepreneurs to amass a fortune by building a business off the Internet.

The former Lockheed systems consultant started McAfee Associates in 1989, initially distributing anti-virus software as "shareware" on Internet bulletin boards.

He took the company public in 1992 and left two years later following accusations that he had hyped the arrival of a virus known as Michelangelo, which turned out to be a dud, to scare computer users into buying his company's products.

McAfee currently has no relationship with the software company, which has since been sold to Intel Corp.

Will Scott Brown run again if John Kerry leaves the Senate? ?We?ll see?

WASHINGTON, D.C.?Massachusetts Republican Sen. Scott Brown returned to Congress this week a lame duck after losing to Democratic challenger Elizabeth Warren. But he could have another chance to keep a place in the Senate if Sen. John Kerry leaves to serve in the Obama administration.

Kerry reportedly is being considered to become President Barack Obama's secretary of defense, and while Brown has declined to announce plans to seek Kerry's seat if it opens, he's not publicly opposing the notion.

"We'll see what happens," Brown told reporters during a press conference on Capitol Hill on Tuesday. "I'm not even thinking about it right now."

Brown was elected in 2010 in a special election after the death of former Sen. Edward Kennedy, who had represented Massachusetts since 1962. Since then, Brown has branded himself as a centrist Republican willing to cross party lines.

On Tuesday, Brown called on Republicans to move closer to the center as a way to broaden the party's appeal.

"We need to be a larger tent party. I'm a pro-choice moderate Republican. There's a vanishing breed here, you know that now," Brown said. "That group in the middle, it's vanishing. And on both sides there are extremes as you all know, kind of pushing against the middle. And I've always felt that group in the middle is quite frankly the most powerful group. I'm hopeful that we'll be a more tolerant, open-minded party."

If Kerry does leave the Senate, Massachusetts will be legally required to hold a special election within 160 days of the seat's vacancy.

Brown also declined to speculate on whether he would run for governor in 2014.

"There is not an opening right now for governor, nor is there an opening for senator," he said. "But there is an opening for a dad and a husband, and that's the role that I want to play."

Egypt's Brotherhood slams Israel over Gaza strikes

CAIRO (AP) ? Egypt's powerful Muslim Brotherhood sharply criticized Israeli leaders on Tuesday over airstrikes in the Gaza Strip, accusing them of heating up the conflict to score political points ahead of elections.

The latest round of violence began Saturday, with rocket attacks from Gaza militants and Israeli airstrikes that killed seven Palestinians. More than 100 rockets have exploded in Israel since the weekend. The exchanges appeared to die down on Tuesday.

Also, Israeli tanks struck a Syrian artillery launcher Monday after a mortar shell flew into Israel-held territory, fueling concerns that Israel could be dragged into the Syrian civil war.

In its statement, the Brotherhood's Freedom and Justice Party referred to Israel as a "Zionist occupier" and a "racist state," placing Israeli Prime Minister Benjamin Netanyahu and Foreign Minister Avigdor Lieberman on the "fringes" of the "far right."

"In the framework of elections that Israel is witnessing is a recent military escalation against occupied Gaza and the occupied Golan Heights," the statement said. Israel has set parliamentary elections for Jan. 22.

The Brotherhood's party called on Arab and Muslim governments "to stop the Zionist war that is operating under electoral calculations for personal gain far from humanitarian calculations for peace, security and stability."

The Muslim Brotherhood itself released a separate statement shortly after its party's, sharpening the criticism and accusing Israel of following a policy that tries to appear opposite itself "and God knows they are liars."

"The killing of tens of our innocent Palestinian brothers is part of a link in a chain of oppression and Judaization that seeks to impose itself on the ground, and that will never materialize with God's will," it said.

The harsh pronouncements followed a small demonstration in Cairo Monday and open letter signed by several liberal parties and revolutionary groups denouncing the Israeli strikes on Gaza.

The statements by both the Brotherhood and its political party highlight decades of tensions between neighbors Israel and Egypt, despite a 1979 peace treaty. The Islamists, repressed in Egypt under the regime that was ousted last year, have emerged as the most powerful group since last year's popular uprising. They won parliamentary elections and the presidency.

Last month, the group's supreme leader, Mohammed Badie, released a fiery tirade against Jews, accusing them of spreading corruption, slaughtering Muslims and desecrating holy sites. The comments were denounced by Israeli officials and a leading anti-Semitic watchdog group.

Unlike his predecessor, the ousted Hosni Mubarak, President Mohammed Morsi has not met Israeli officials since his election in June. He has also not mentioned Israel by name in official statements, in line with longstanding Brotherhood policy.

But to secure investments and bolster the economy, Morsi recently met with U.S. business executives from top American companies and vowed to respect his country's peace accord with Israel.

Since his election, Israel and Egypt have quietly cooperated over a military buildup in the Sinai Peninsula, where Islamist militants have been attacking Egyptian forces and launching attacks into Israel. The security coordination signals there is no significant shift in Cairo's policies toward Israel, despite the heated Brotherhood rhetoric.

Morsi has also indicated there will not be radical changes in policy anytime soon, even with Hamas, an offshoot of the Muslim Brotherhood movement, which rules the Gaza Strip,.

Morsi's government still tightly controls Palestinian movement through the Rafah border crossing with Gaza. Egypt's role in a blockade of Gaza, imposed by Israel after Hamas won elections and later overran the territory in 2007, is highly unpopular.

Israel withdrew from Gaza in 2005 but controls its airspace, seacoast and most land crossings.

In a meeting in Cairo Tuesday with Western-backed Palestinian President Mahmoud Abbas, Morsi expressed his "full support" for Palestinian plans to seek nonmember state status at the United Nations.

Israel and the U.S. oppose the move.

Clouds may ruin trip for eclipse fans in Australia

SYDNEY (AP) ? Tens of thousands of tourists, scientists and amateur astronomers who traveled from around the world to see a total solar eclipse in northern Australia may be getting shortchanged by the weather.

Forecasters were predicting cloudy skies around dawn Wednesday, when the moon will pass between the sun and Earth and plunge a slice of Australia's northeast into darkness. Many worried that they will miss a rare chance to view the celestial phenomenon.

"There will be breaks in (the clouds), but it's just a matter of the luck of the draw whether you get a break at the right time," said Queensland state Bureau of Meteorology forecaster Andrew Mostyn. "It's a bit of bad luck."

The eclipse will cast its 150-kilometer (95-mile) wide shadow starting at dawn in Australia's Northern Territory and then cross the northeast tip of the country before swooping east across the South Pacific. No islands are in its direct path, so northern Australia is the only land where there's even a chance of seeing the full eclipse, said Geoff Wyatt, an astronomer with Sydney Observatory.

A partial eclipse will be visible from east Indonesia, the eastern half of Australia, New Zealand, Papua New Guinea and southern parts of Chile and Argentina. Totality ? the darkness that happens at the peak of the eclipse ? will last just over two minutes.

Among those sweating out the forecast was U.S. astronomer Jay Pasachoff, who traveled to Australia in hopes of viewing his 56th solar eclipse. Pasachoff, an astronomy professor at Williams College in Massachusetts, and a team of about 50 scientists and students have fanned out across the region to improve the odds that at least some of them will see the eclipse. The group is planning to study the sun's corona, the glowing white ring around the sun that is visible only during an eclipse.

Despite the anxiety over the weather and the long journey to get there, Pasachoff said he wouldn't miss it.

"Just imagine you were a heart surgeon and someone actually told you you could look inside a human heart only for two minutes, and only if you went halfway around the world," he said. "You would do it."

Some Queensland hotels have been booked up for more than three years and more than 50,000 people have flooded into the region to watch the solar spectacle, said Jeff Gillies, regional director of Queensland Tourism.

Skygazers are planning to crowd beaches, boats, fields and hot air balloons to watch the event. Fitness fanatics will race in the Solar Eclipse Marathon, where the first rays of the sun re-emerging from behind the moon will serve as the starting gun. Some have already been partying for days at a weeklong eclipse festival.

Scientists will be studying how animals respond to the eclipse, with underwater cameras capturing the effects of sudden darkness on the creatures of the Great Barrier Reef.

"It's an unknown with how they'll react," Gillies said. "A little bit of flora and fauna confusion, I would imagine."

The last total solar eclipse visible in Australia was 10 years ago, in the South Australia Outback.
